Peptide Reconstitution & Storage Guide

A rigorous analytical protocol covering aseptic handling, vacuum management, reconstitution calculation matrices, and optimal thermal preservation for laboratory peptides.

Research Use Only (RUO) Compliance Notice
This laboratory protocol is compiled strictly for analytical chemists, biochemists, and academic researchers conducting in-vitro scientific investigations. Apex Pure Pep synthesized biochemicals are strictly prohibited from in-vivo human administration, clinical trial, or veterinary consumption.

1 Essential Equipment & Consumables Checklist

Prior to breaking seals, verify that all necessary sterile items are arranged inside a certified laminar flow hood or a sanitized clean-air workstation:

Lyophilized Peptide Vial

Unopened Apex Pure Pep lyophilized powder vial, inspected for intact vacuum seal and butyl stopper.

Bacteriostatic Water (BAC)

Sterile water containing 0.9% (9 mg/mL) Benzyl Alcohol USP as a multi-dose antimicrobial preservative.

Sterile Syringes & Needles

U-100 insulin syringes (0.5 mL or 1.0 mL with 30G/31G needles) or separate 21G–25G reconstitution syringes.

Antiseptic Isopropyl Wipes

Individually wrapped 70% Isopropyl Alcohol (IPA) pads for septum and vial rim disinfection.

Sharps Disposal Container

Designated puncture-resistant biohazard bin for safe containment of contaminated sharps.

Light-Protective Secondary Box

Opaque vial rack, amber secondary tube, or aluminum foil wrap for light-sensitive peptides.

2 Reconstitution Concentration Mathematical Matrix

Accurate laboratory volumetric dosing requires standardized concentration calculations. Do not guess solvent volume; use the direct mass-to-volume formula below:

Concentration (mg/mL) = Total Peptide Mass (mg) ÷ Added Diluent Volume (mL)
Concentration per Syringe Unit (μg/unit) = [Total Mass (mg) × 1,000] ÷ [Diluent (mL) × 100 units]
Note: In standard U-100 insulin syringes, 1.0 mL = 100 graduation units (0.01 mL per graduation mark). For example, a 5 mg vial dissolved in 2.0 mL of BAC water yields 2.5 mg/mL, which translates to 25 μg per 1 syringe unit.

Peptide Vial Mass BAC Water Volume Final Concentration (mg/mL) Micrograms per U-100 Unit Recommended Usage Case
2 mg 1.0 mL 2.0 mg/mL 20 μg / unit High-precision low-dose assays
5 mg 1.0 mL 5.0 mg/mL 50 μg / unit Standard laboratory titer
5 mg 2.0 mL 2.5 mg/mL 25 μg / unit Easy volumetric titrations
10 mg 2.0 mL 5.0 mg/mL 50 μg / unit High-throughput research assays
10 mg 3.0 mL 3.33 mg/mL 33.3 μg / unit Extended stability dilution
15 mg 3.0 mL 5.0 mg/mL 50 μg / unit Standardized 50μg/unit metric

3 Step-by-Step Aseptic Reconstitution Protocol

1

Thermal Equilibration & Septum Disinfection

Remove the peptide vial from deep freezer (-20°C) and allow it to sit on the benchtop for 15 to 20 minutes until it equilibrates to room ambient temperature. This prevents atmospheric moisture condensation on the cold cake inside.

Pop off the flip-off protective cap to expose the butyl rubber stopper. Firmly wipe the septum using a fresh 70% Isopropyl Alcohol prep wipe.

Crucial Rule: Allow the rubber stopper to air-dry completely for 20 to 30 seconds before needle insertion. Puncturing wet rubber pushes trace alcohol inside the vial, which can denature sensitive peptide chains.

2

Diluent Draw & Vacuum Pressure Equalization

Using a sterile syringe, draw the calculated volume of Bacteriostatic Water. Ensure no air pockets remain in the syringe barrel.

Factory Vacuum Awareness: Apex Pure Pep vials are sealed under negative atmospheric pressure (factory vacuum) to protect against oxidation. When inserting the needle, the internal vacuum will naturally pull the syringe plunger inward.

  • Hold plunger firmly with your thumb: Do NOT allow the diluent to be violently sucked into the vial by unregulated vacuum force.
  • Sudden high-velocity impact can physically crush and shear delicate tertiary peptide conformations.

3

Gentle Sidewall Injection Technique

Angle the needle at approximately 45 degrees toward the inner glass wall of the vial, keeping the tip well above the freeze-dried cake.

Slowly and smoothly depress the plunger, allowing the diluent stream to trickle down the interior glass wall. The solvent should pool at the bottom of the vial and slowly submerge the lyophilized cake from beneath.

Strict Prohibition: NEVER blast diluent directly onto the center of the lyophilized cake. Direct pressure causes structural degradation and foaming.

4

The Absolute “NO-SHAKING” Rule & Dissolution

Once the solvent is added, withdraw the needle safely into your sharps container. Peptides are fragile polymer chains of amino acids; mechanical turbulence breaks their spatial structure.

  • NEVER shake, vortex, or aggressively invert the vial. Shaking generates foam and causes irreversible molecular aggregation (precipitation).
  • Hold the vial between your fingertips and swirl in slow, gentle horizontal circles, or roll it softly between your palms.
  • Allow the vial to rest undisturbed at room temperature for 5 to 10 minutes. High-purity peptides will dissolve completely into a crystal-clear, transparent solution.

4 Storage Protocols, Temperature Zones & Stability

Lyophilized Powder (Dry Cake)
-20°C ± 5°C

Primary Storage Zone: Deep Freezer

Shelf Stability: 24 to 36 Months

Short-Term Transit: Ambient (up to 37°C) stable for 30 days without loss of potency.

Keep in original sealed amber/clear vials away from light and ambient moisture.

Reconstituted Solution (Liquid)
2°C to 8°C

Primary Storage Zone: Standard Refrigerator

Shelf Stability (BAC Water): 21 to 30 Days

Shelf Stability (Plain Sterile Water): 24 to 48 Hours maximum

Never store in door trays where temperature fluctuates upon opening.

CRITICAL WARNING: Never Freeze Reconstituted Solution in Frost-Free Freezers
Once reconstituted into liquid form, DO NOT place the vial in a domestic or standard laboratory frost-free freezer. The automated auto-defrost warming cycles generate repeated micro-freeze/thaw episodes that create ice crystal shear planes, permanently cleaving peptide bonds and rendering the compound inactive. If freezing a reconstituted solution is strictly necessary, aliquot into single-use cryogenic vials and flash-freeze at -80°C (thaw only once; discard residual).

5 Laboratory Troubleshooting & Inspection Matrix

Observation Probable Cause Laboratory Corrective Action
Turbid / Cloudy Solution Incomplete dissolution, hydrophobic peptide sequence, or aggressive vortexing. Allow the vial to sit stationary at room temperature for 15 minutes. For hydrophobic sequences, adjust pH with trace sterile acetic acid (0.1%) or DMSO as specified in product data sheet.
Microscopic Foaming Vial was agitated or solvent was injected with excessive velocity directly on cake. Do not agitate further. Refrigerate at 4°C undisturbed for 30–60 minutes until micro-bubbles completely collapse into solution.
Absence of Vacuum Draw Septum micro-fissure or standard atmospheric pressure filling. Inspect aluminum crimp. If seal is intact, verify mass using analytical balance before reconstitution. Report lot number to Apex Pure Pep QA team if defect is suspected.
Rubber Core Particulate Blunt or perpendicular needle puncture fragmented the butyl rubber stopper. Always insert needles at a 45°ā€“60° angle with the needle bevel facing upwards, then gently straighten to 90° during penetration. Discard if rubber particulate is observed in solution.
